No, Hamilton and Burr were close for a very little period, they study law together in New York. Their conflict began when Burr made Hamilton angry when he won against Hamilton father in law in senate election. Their conflict was continue in 1800, when Burr had announced that he was going for president race, Hamilton insulted him and stood agianst him supporting Thomas Jefferson. The challenge was continue till 1804 when Burr invited hamilton for a duel, Burr had shot hamilton and made him dead.
